Genetic diversity of Neotyphodium fungal endophytes in three Iranian grass species using AFLP molecular markers
Authors
Abstract:
Genetic diversity of fungal endophytes, Neotyphodium species, was studied in grasses Festuca arundinacea, F. pratensis and Lolium perenne using AFLP markers. Fungi were isolated from the host leaf sheaths and Neotyphodium species were selected based on morphological characteristics. To confirm identity of selected fungi belonging to the genus Neotyphodium, polymerase chain reaction was performed using specific primers. AFLP with eight primer combinations was conducted to assess genetic variation for endophyte isolates. Cluster analysis based on AFLP data placed isolates into three separate groups according to their host species.

Genetic diversity of Iranian melon cultigens revealed by AFLP markers
Iran is a part of the secondary centre of origin of melons, and therefore, there is a wealth of genetic variation of these species in this country. The Amplified Fragment Length Polymorphism (AFLP) marker was applied to investigate the genetic variations among five major Iranian melon cultigens (Cucumis melo) and ‘Ananasi’ as a general well-known cultivar. Ten primer pairs were used on 90 indiv...
full textAssessment of Genetic Diversity in Some Iranian Populations of Bunium persicum Using RAPD and AFLP Markers
The genetic diversity of 20 Iranian populations of Bunium persicum has been evaluated with random amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP) markers. Fresh leaves of seedlings from each population were used for genomic DNA extraction. Analysis of banding patterns of 15 RAPD primers and 17 AFLP primer combinations, revealed 192 (86%) and 228 (75%) polymorp...
full textAssessment of SCoT and DAMD molecular markers in genetic diversity and species delimitation of three moss species grown in Iran
This study was conducted to assess the efficacy of SCoT and DAMD molecular markers in genetic diversity of three moss species and compare them with ISSR marker. Another objective was to evaluate the suitability of these DNA markers in species identification (delimitation) in three moss species, including Neckera complanata, Homalothecium sericeum and Neckera cris...
full textGenetic diversity among tall fescue ecotypes using agro-morphological traits, nutritional values and AFLP molecular markers
This study was carried out to compare agro-morphological traits and nutritive value of 25 tall fescue (Festuca arundinacea) ecotypes grown in Tehran, Iran. The experiments were carried out in a split plot design during 2016-2018 growing seasons. The results showed that there was a significant genetic variation in existing germplasm. The highest heritability was related to dry forage yi...
full textEvaluation of Genetic Diversity in Iranian Violet (Viola spp) Populations Using Morphological and RAPD Molecular Markers
Recognition of genetic reserves and desirable genes is the basis of breeding programs. So far, in Iran, due to the lack of recognition of genetic resources, a considerable breeding program has not been done on native plants. The study of the genetic diversity of violets as a native plant with ornamental and medicinal uses is the great importance in advancing the breeding goals of this plant. So...
full textEcological importance of Neotyphodium spp. grass endophytes in agroecosystems
Neotyphodium spp. fungal endophytes form symbiotic associations with agronomic grasses with expression ranging from mutualistic to parasitic. In general, endophyte infection frequencies seem more variable in natural compared to pastoral situations, suggesting that expression and benefits attributable to endophyte infection depend on the resource environment in which the host–endophyte complex w...
